Genomic alterations drive tumor heterogeneity and therapeutic resistance, yet capturing these dynamics at single-cell resolution remains challenging. This session explores cutting-edge strategies integrating Oxford Nanopore’s multiomic sequencing with single-cell technologies to uncover novel cancer biology. We highlight a multiomic approach tracking clonal evolution in B-ALL and a novel method, ecSingle, revealing the role of extrachromosomal DNA (ecDNA) in tumor progression. Together, these insights provide a comprehensive and simultaneous view of genomic, epigenomic, and transcriptomic information across thousands of single cells linking genomic alterations to functional consequences.
